Stains used in biology

I saw alot of quesitions in the ATP, about the preparations of cells, i know how to do that but could you please tell me which stains to use and when. Also how to prepare pollen grains for study is not in my book would someone help me with that too ? :) thanks.

Methylene Blue and Iodine Solution are common stains.
For pollen grain:
Gently rub the grains from part of flower so that they fall onto the glass slide. First keep them in water to prevent their dehydration. Then put them on the slide and add a suitable stain. put the coverslip while preventing the formation of any air bubbles. Dry any excess liquid by using a blotting paper.
Put the slide under a microscope. Set the magnification and light intensity and observe. :)
